Goals / Objectives
To inoculate citrus germplasm with the bacteria responsible for citrus huanglongbing (HLB) disease in order to evaluate germplasm for resistance to the disease.
Methods (unparsed)

USDA-ARS-U.S. Horticultural Research Laboratory, Fort Pierce Florida, is producing thousands of citrus scion or rootstock plants transformed to express peptides that might mitigate citrus greening disease; additional plants are being continually produced. The more rapidly this germplasm can be evaluated, the sooner we will be able to identify transgenic strategies for controlling the disease. The purpose of this proposal is to support a high-throughput inoculation program for the purposes of infecting and evaluating transgenic citrus for hunaglongbing resistance. Briefly, individual plants are caged with infected psyllids for two weeks, and then housed for six months in a greenhouse with an open infestation of infected psyllids. Plants are then moved into a psyllid-free greenhouse and evaluated for growth, huanglongbing symptoms and Liberibacter asia ticus titer, and later to the field for further evaluations. The funds requested will be for two technicians dedicated to the project, primers and associated supplies for PCR, and general supplies including insect cages.
