Goals / Objectives
The purpose of this project is to apply cryotherapy and the use of 'mini-plant-indexing' to accelerate the cleaning of germplasm accessions for use in citrus breeding in Florida. We have demonstrated the effectiveness in using cryotherapy to eliminate HLB, citrus viroids, and Citrus tatterleaf virus (pathogens which are the most difficult to eliminate using traditional shoot tip grafting). This is a request for funds for technical support to clean priority materials for distribution and use in the Florida Citrus breeding programs.
Methods (unparsed)

Year 1. In Colorado, 30 accessions from Riverside ARS will be processed for cryotherapy. Plants are grown in vitro on seedling rootstocks for 15 weeks, sent to Riverside ARS, and then tested using established laboratory based diagnostics for HLB and other graft transmissible pathogens. In vitro clones testing negative will be established in quarantine in California (under permit). Testing will continue using young indicator plants for biological indexing. It is anticipated that 15 of the accessions will be released from quarantine at the end of year 1. In year 2, an addition 45 plants will be processed for cryotherapy, and subsequently indexed by laboratory and biological indexing methods for release.
